肠杆菌属细菌一类整合子及其基因盒特征

整合子是捕获、整合和表达外源基因的重要元件,可以加速抗生素抗性基因的传播。本研究分析了已完成全基因组测序肠杆菌属细菌一类整合子及其基因盒的存在情况与特征。实验表明,目前62个肠杆菌属细菌,包括40个阴沟肠杆菌、11个霍氏肠杆菌和11个其它肠杆菌都完成了全基因组测序,其中40.32%(25/62)肠杆菌属细菌含有一类整合子。在25个含一类整合子的肠杆菌中,72%(18/25)携带一个整合子,28%(7/25)含有多个整合子。25个肠杆菌共存在36个整合子,其中88.89%(32/36)位于质粒上,其余11.11%(4/36)位于染色体上。肠杆菌属细菌一类整合子整合酶基因(1 014 bp)高度保守,其中一个整合子整合酶基因发生了3碱基突变(3/1 014),7个发生2碱基突变(2/1 014),13个发生1碱基突变(1/1 014 bp),其余没有突变。25个含一类整合子肠杆菌共存在18种基因盒,其中dfrA1基因盒频率最高16.13%(10/62),其次是aadA1(8.06%, 5/62),接着是aacA27-ereA-IS1247-aac3-arr2-ereA基因盒(4.84%, 3/62),其他基因盒频率低于4.0%。本研究有助于了解一类整合子在肠杆菌属细菌产生抗生素抗药性中的重要作用。

Integron, which could accelerate the spread of antibiotic resistance genes, is an important element to capture, integrate and express exogenous genes. The presence and characteristics of class 1 integrons as well as gene cassettes were investigated in Enterobacteria that have completed the whole genome sequencing. The results showed that currently 62 Enterobacteria including 40 Enterobacter cloacae, 11 Enterobacter hormaechei and 11 others which have completed the whole genome sequencing, 40.32%(25/62) of which contains class 1 integrons.Among 25 Enterobacteria, 72%(18/25) carried a single integron and 28%(7/25) contained more than one integron.88.89%(32/36) of integrons are locatedon plasmids and the rest 11.11%(4/36) are found in chromosomes among the 36 integrons that distributed in 25 Enterobacteria, The class 1 integron-integrase gene(1 014 bp) in Enterobacteria are highly conserved, of which one had three base mutations(3/1 014), seven had two base mutations(2/1 014),13 had one base mutation(1/1 014), and the rest had no mutation. A total of 18 gene cassettes are present in 25 enterobacter species containing class 1 integron, of which dfrA1 gene cassette has the highest frequency(16.13%,10/62), followed by aadA1(8.06%, 5/62), and then aacA27-ereA-IS1247-aac3-arr2-ereA(4.84%, 3/62). The frequency of others gene cassette was less than 4.0%. This study was helpful to understand the important role of class 1 integron in the emergence of antibiotic-resistant Enterobacteria.
