FAD2是亚油酸合成的关键酶,是控制高油酸特性的重要基因。本研究采用RT-PCR技术在蒜头果中成功克隆得到一个FAD2基因,命名为MoFAD2,GenBank登录号为MK210593。其含有1 329 bp开放阅读框,编码442个氨基酸。序列比对分析显示MoFAD2拥有FAD2家族特有的3个保守的组氨酸中心位点,蒜头果FAD2与中粒咖啡(Coffea canephora) FAD2的蛋白序列同源性为80.00%。系统进化树分析显示MoFAD2与石榴(Punica granatum)、珙桐(Davidia involucrata)聚为一支。荧光定量PCR分析表明MoFAD2在果实和叶中均有表达,但在蒜头果果实膨大期的表达量最高。研究为将来更好地开发利用蒜头果油用价值和进一步开展FAD2在不饱和脂肪酸生物合成过程中的调控作用研究奠定基础。

omega-6 fatty acid desaturase is the key enzyme in the biosynthesis pathway of linolenic acids. It is an important gene controlling the characteristics of high oleic acid. In this study, aω-6 fatty acid dehydrogenase gene(MoFAD2) was obtained successfully by cloned using RT-PCR technology from Malania oleifera. The gene sequence of MoFAD2 consisted of 1 329 bp open reading frame(ORF) and encoded 442 amino acids, accession number of the MoFAD2 gene in GenBank was MK210593. The proteinic sequence alignment revealed there is 80.00% of similarity between Malania oleifera and Coffea canephora. The phylogenetic tree analysis showed that MoFAD2 and FAD2 from Punica granatum and Davidia involucrata had the same branch, and has 3 domains functionally conserved that is special in the FAD2 family. The fluorescent quantitative reverse PCR certified that the expression of MoFAD2 gene in fruit of the flower withered 3 months is highest. The study laid a foundation for better development and utilization of Malania oleifera fruit oil and further studies on the regulatory role of FAD2 in the biosynthesis of unsaturated fatty acids in the future.
